Marcos Pereira, Elis Eleutherio, Anita Panek; Acquisition of tolerance against oxidative damage in Saccharomyces cerevisiae: (in English) AbstractBackgroundLiving cells constantly sense and adapt to redox shifts by the induction of genes whose products act to maintain the cellular redox environment. In the eukaryote Saccharomyces cerevisiae, while stationary cells possess a degree of constitutive resistance towards oxidants, treatment of exponential phase cultures with sub-lethal stresses can lead to the transient induction of protection against subsequent lethal oxidant conditions. The sensors of oxidative stress and the corresponding transcription factors that activate gene expression under these conditions have not yet been completely identified.ResultsWe report the role of SOD1, SOD2 and TPS1 genes (which encode the cytoplasmic Cu/Zn-superoxide dismutase, the mitochondrial Mn-isoform and trehalose-6-phosphate synthase, respectively) in the development of resistance to oxidative stress. In all experimental conditions, the cultures were divided into two parts, one was immediately submitted to severe stress (namely: exposure to H2O2, heat shock or ethanol stress) while the other was initially adapted to 40°C for 60 min. The deficiency in trehalose synthesis did not impair the acquisition of tolerance to H2O2, but this disaccharide played an essential role in tolerance against heat and ethanol stresses. We also verified that the presence of only one Sodp isoform was sufficient to improve cellular resistance to 5 mM H2O2. On the other hand, while the lack of Sod2p caused high cell sensitivity to ethanol and heat shock, the absence of Sod1p seemed to be beneficial to the process of acquisition of tolerance to these adverse conditions. The increase in oxidation-dependent fluorescence of crude extracts of sod1 mutant cells upon incubation at 40°C was approximately 2-fold higher than in sod2 and control strain extracts. Furthermore, in Western blots, we observed that sod mutants showed a different pattern of Hsp104p and Hsp26p expression also different from that in their control strain.ConclusionsTrehalose seemed not to be essential in the acquisition of tolerance to H2O2 stress, but its absence was strongly felt under water stress conditions such as heat and alcoholic stresses. On the other hand, Sod1p could be involved in the control of ROS production; these reactive molecules could signal the induction of genes implicated within cell tolerance to heat and ethanol.   1. Okuno, H., Osato, N., Takeuchi, I.; Firmware Approach to Fast Lisp Interpreter: The approach to speed up a Lisp interpreter by implementing it in firmware seems promising. A microcoded Lisp interpreter shows good performance for very simple benchmarks, while it often fails to provide good performance for larger benchmarks and applications unless speedup techniques are devised for it. This was the case for the TAO/ELIS system. This paper describes various techniques devised for the TAO/ELIS system in order to speed up the interpreter of the TAO language implemented on the ELIS Lisp machine. The techniques include data type dispatch, variable access, function call and so on. TAO is not only upward compatible with Common Lisp, but also incorporates logic programming, object-oriented programming and Fortran/C-like programming into Lisp programming. TAO also provides concurrent programming and supports multiple users (up to eight users). The TAO interpreter for those programming paradigms is coded fully in microcodes. In spite of rich functionalities, the speed of interpreted codes of TAO is comparable to that of compiled codes of commercial Lisp machines. Furthermore, the speeds of the interpreted codes of the same program written in various prograrnming paradigms in TAO does not differ so much. This speed balance is very important for the user. Another outstanding feature of the TAO/ELIS system is its firmware development environments. Micro Assembler and Linker are written in TAO, which enables the user to use the capability of TAO in microcodes. Since debugging tools are also written in mini-Lisp, many new tools were developed in parallel to debugging of microcodes. This high level approach to firmware development environments is very important to provide high productivity of development. [Text] (1.85)

Mittels RFA, Coulometrie, ICP-MS und weiterer Methoden wurden ca. 550 Proben zweier Kernbohrungen (Kirchrode II/94, Hoheneggelsen KB40) aus dem Raum Hannover auf ihre Haupt- und Spurenelementzusammensetzung (u.a. REE, As, Se, Cd) und teilweise auch auf Schwefel-Isotopenverhaltnisse hin analysiert. Zusatzlich wurden an einigen Proben Tonmineralanalysen durchgefuhrt. Ziel der Untersuchungen ist es, Aussagen uber die damaligen Ablagerungsbedingungen wie die Bioproduktivitat im Oberflachenwasser, Redoxbedingungen in der Wassersaule, an der Sediment-Meerwasser-Grenze und im Sediment oder anderungen des detritischen Elementeintrags zu treffen. Aufgrund einer detaillierten Bearbeitung einiger Teilabschnitte der Profile (Probenabstande kleiner 1cm) werden zudem kleinskalige Variationen der Elementgehalte innerhalb einzelner Sedimentabschnitte sowie im cbergangsbereich von sub- zu anoxisch bzw. anoxisch zu oxisch verdeutlicht. In this thesis results of high-resolution geochemical investigations on sediments of the boreal Lower Cretaceous (Apt/Alb) from north western Germany are presented. About 550 samples taken from two drill cores (Kirchrode II/94, Hoheneggelsen KB40) out of the Hannover area were measured by XRF, coulometry, ICP-MS and other methods for their major and minor elemental composition (e.g. REE, As, Se, Cd). On several samples the sulfur isotopic ratios were determined too. Additionally on some selected samples the clay mineralogy has been analysed. The aim of this study is a contribution to the understanding of the former depositional conditions, bioproductivity in surface waters and redox conditions in the water column, at the sediment-water interface and in the sediment itself. The detailed analysis of some special core segments (sampling intervals < 1 cm) allows the detection of small-scale variations in element contents in individual sedimentary units and in transitional stages between suboxic to anoxic and anoxic to oxic areas. Tankel, Jesslyn Elise; A Teaching Toy: Free Forms in an Abstract Landscape: Anna Marshall-Baker, Mitzi Vernon, Richard Goff, Ellen Braaten; (in English) As a young student, I enjoyed a passion for the studio arts and an interest in art history. I ardently pursued these areas in college, continuing their exploration in graduate school in the field of Industrial Design. For the thesis project, my objective is to foster in children an appreciation for art and, possibly, to help them develop a critical and appreciative eye toward their environment. In my research, I find inspiration in child psychology and the toys of childs play, in 15th-century Islamic art and 20th-century Abstraction, and in naturally occurring events such as ivy twisting up a knotty tree trunk and soft ripples in a pond. <p> The assimilation of these elements, together with the ideas that they generate, evolve into the foundation for my project. My intent is to design a toy which provides children ages six to twelve years with insight into both the world of nature and the realm of art. Further, the toy will heighten each childs visual vocabulary to acknowledge the organic, natural line that exists in nature and which is, perhaps, represented in abstract art.
